Retatrutide (LY3437943) is an investigational once-weekly triple hormone-receptor agonist engineered by Eli Lilly. It is the first molecule to combine agonism at all three incretin-related receptors — GLP-1, GIP, and glucagon — in a single 39-amino-acid backbone stabilized by a C20 fatty-diacid side chain that extends plasma half-life to ~6 days.
